Reverse Transcription Polymerase Chain Reaction is a technique which is responsible for detecting and quantifying the RNA which has been extracted from even a single cell. It is a very sensitive technique as it requires a minute quantity of the sample. On the contrary, there are two more techniques i.e., Northern blot and nuclease protection assays which are widely used but require large amounts of RNA.
One major problem that arises in RT-PCR is that the genomic DNA gets mixed up with RNA which leads to incorrect results. This reaction is catalyzed by an enzyme which is known as reverse transcriptase. It uses RNA as a template and forms complementary DNA which is not prone to degradation by RNase.
